Optical tweezers is one of the most effective methods for manipulating micron and sub-micron sized particles non- destructively. In this paper a tapered fiber optical tweezers is calibrated using power spectrum analysis of the trapped particle motions. We utilized a high- speed CMOS camera with 500 frames per second for particle position detection. As validation experiment, using this setup, stretching of a multilamellar lipid bilayers is demonstrated.
