In this works a lightguide-based total internal reflection fluorescence microscope is designed and demonstrated to detect the fluorescence of fluorophore molecules labeled in the sample. Total internal reflection fluorescence microscopy (TIRFM) is a technique that utilizes the total internal reflection phenomenon. Indeed, when total internal reflection occurs, an evanescent wave is generated in the less dense medium which is used for illuminating the sample and consequently the possibility of observing a superficial (instead of bulk) part of fluorophore-labeled sample is opened up. For this purpose, an inverted microscope first has been designed using the available optic and optomechanical devices. Subsequently, the arrangement of lightguide-based total internal reflection fluorescence microscope has been done by means of the inverted microscope to carry out the imaging operation for some CdSe quantum dot samples
